Ipamorelin / CJC-1295 (no DAC) is a blend of two synthetic peptides in one vial, also listed as CJC / Ipa: ipamorelin, a five-residue agonist of the ghrelin receptor, and CJC-1295 without DAC, a 29-residue analog of growth hormone-releasing hormone. Each component has its own literature, and each acts on a different receptor of the pituitary somatotroph. No published study of the blend itself was found.
Supplied for laboratory research use only. Not for human or veterinary use, and not for diagnostic or therapeutic application.
About Ipamorelin / CJC-1295 (no DAC)
Structure of Ipamorelin / CJC-1295 (no DAC)
The vial holds 5 mg of each component. Ipamorelin is the pentapeptide Aib-His-D-2-Nal-D-Phe-Lys-NH2. CJC-1295 without DAC is the 29-residue amide Tyr-D-Ala-Asp-Ala-Ile-Phe-Thr-Gln-Ser-Tyr-Arg-Lys-Val-Leu-Ala-Gln-Leu-Ser-Ala-Arg-Lys-Leu-Leu-Gln-Asp-Ile-Leu-Ser-Arg-NH2, a sequence PubChem lists among the names on its CJC-1295 record. The compound that the 2005 paper named CJC-1295 has one more residue: a C-terminal lysine carrying a maleimidopropionyl group, the part later called the Drug Affinity Complex, or DAC. The no-DAC form lacks that lysine and group. Being a mixture, the blend has no single formula, weight or registry number.
Where Ipamorelin / CJC-1295 (no DAC) comes from
The two components come from separate programs. Raun and colleagues at Novo Nordisk in Måløv, Denmark, described ipamorelin in 1998, within a series of compounds lacking the central Ala-Trp dipeptide of growth hormone-releasing peptide 1. Jetté and colleagues at ConjuChem in Montreal described CJC-1295 in 2005 as a tetrasubstituted form of human growth hormone-releasing factor (1-29) with an added maleimide-bearing lysine, designed to bond to the free thiol on cysteine 34 of serum albumin. The peptide without that lysine goes by CJC-1295 no DAC. The pairing of the two is a supplier convention, not a compound from the literature.
What Ipamorelin / CJC-1295 (no DAC) binds
The components reach the same pituitary cell through different receptors. The 1998 study showed with receptor antagonists that ipamorelin stimulates growth hormone release through a GHRP-like receptor, which UniProt lists as growth hormone secretagogue receptor type 1, coupled to Gq proteins and phospholipase C. CJC-1295 was shown to activate the growth hormone-releasing factor receptor on rat anterior pituitary cells; UniProt describes that receptor as coupled to G proteins that activate adenylyl cyclase. Those data were obtained with the DAC form bound to albumin. How the two peptides behave when present together has not been reported.
Ipamorelin / CJC-1295 (no DAC) in the research literature
Ipamorelin was characterized in primary rat pituitary cells, anesthetized rats and conscious swine, where plasma growth hormone, ACTH and cortisol were measured. CJC-1295 with DAC was examined as an albumin conjugate for stability against dipeptidylpeptidase-IV in vitro, in a growth hormone secretion assay in cultured rat anterior pituitary cells, and in Sprague Dawley rats, where Western blotting found it on the serum albumin band. Published data on the no-DAC peptide alone are sparse, and the sources consulted contain no study of the two-peptide blend, in cells, animals or people.
Regulatory status of Ipamorelin / CJC-1295 (no DAC)
Neither component is described in these sources as an approved drug, and the blend is not an approved product. FDA lists ipamorelin acetate in Category 2 of its interim policy for bulk substances nominated under section 503B, added September 29, 2023, and lists CJC-1295 among nominations withdrawn by the nominators, in each case citing a possible risk of immunogenicity from peptide-related impurities. The FDA entry does not distinguish the DAC and no-DAC forms. The blend is supplied here as a reference reagent for in-vitro laboratory research only, not for diagnostic or therapeutic application.
Handling Ipamorelin / CJC-1295 (no DAC) in the laboratory
The material is a lyophilized powder in a vacuum-sealed vial. Keep the sealed vial frozen at -4 °F and protected from light, and let it reach room temperature before breaking the seal so that moisture does not condense on the powder. Once in solution, keep it at 39 °F and plan to use it within about four weeks. Divide a solution into single-use aliquots instead of freezing and thawing one container repeatedly. No solubility or stability figure specific to this blend appears in the sources consulted, so none is given here. Because the two peptides differ several-fold in size, equal masses in the vial are not equal molar amounts.

Ipamorelin / CJC-1295 (no DAC): common questions
Is CJC / Ipa the same as Ipamorelin / CJC-1295 (no DAC)?
Yes. CJC / Ipa is another name for Ipamorelin / CJC-1295 (no DAC), which is also written CJC 1295 Ipamorelin. It is the same compound, supplied here as a lyophilized powder for in-vitro laboratory research use only.
What is Ipamorelin / CJC-1295 (no DAC) supplied as?
Ipamorelin / CJC-1295 (no DAC) is supplied as a lyophilized (freeze-dried) powder in a vacuum-sealed vial, in 5 mg / 5 mg, for in-vitro laboratory research use only.
Is Ipamorelin / CJC-1295 (no DAC) available as a 10-vial kit?
Yes. Ipamorelin / CJC-1295 (no DAC) comes in a 10-vial kit: 5 mg / 5 mg for $243.00 ($24.30 per vial), presale prices. A kit is ten sealed vials of the same size, priced below ten single vials.
When do Ipamorelin / CJC-1295 (no DAC) presale kits ship?

Is Ipamorelin / CJC-1295 (no DAC) for human use?
No. It is supplied as a reference reagent for laboratory research use. Not for human or veterinary use, and not for diagnostic or therapeutic application.
What is in the Ipamorelin / CJC-1295 (no DAC) blend?
Two synthetic peptides in one lyophilized vial, 5 mg of each. Ipamorelin is the pentapeptide Aib-His-D-2-Nal-D-Phe-Lys-NH2. CJC-1295 without DAC is a 29-residue amide derived from growth hormone-releasing factor (1-29), with D-alanine at position 2, glutamine at 8, alanine at 15 and leucine at 27.
What does no DAC mean for CJC-1295?
DAC stands for Drug Affinity Complex. In the compound published as CJC-1295 it is an added C-terminal lysine carrying a maleimide group that bonds to the free thiol on cysteine 34 of serum albumin. The no-DAC peptide is the same 29-residue chain without that lysine, so it has no albumin-reactive group.
Do ipamorelin and CJC-1295 bind different receptors?
Yes. Ipamorelin acts at the growth hormone secretagogue receptor, the receptor for ghrelin, which couples to Gq and phospholipase C. CJC-1295 is an analog of growth hormone-releasing factor and activates that hormone's receptor, which UniProt describes as coupled to G proteins that activate adenylyl cyclase.
